- 英文摘要: The aim of this present study is to investigatethe impacts of combinatorial simvastatin administrationand endothelial progenitor cell (EPC) transplantation ontherapeutic angiogenesis in an athymic nude mouse modelof hind limb ischemia. Athymic nude mice were dividedinto four groups (n = 10/group): vehicle administrationplus PBS injection (control), simvastatin administrationplus PBS injection (simvastatin), vehicle administrationplus EPC transplantation (EPC), and simvastatin administrationplus EPC transplantation (combination). Thecombination therapy had the greatest laser Doppler bloodperfusion imager (LDPI) index and capillary densityamong the four groups. Importantly, this combinationtherapy significantly reduced apoptosis of ischemic skeletalmuscle cells in part through downregulation of Bax andupregulation of Bcl-2 compared with the other groups.Moreover, the combination therapy exhibited the highestefficacy of increasing the ratio of phospho-Akt to Aktamong the four groups. Taken together, the simvastatin andEPC combination therapy promotes powerful angiogenesisin hindlimb ischemia. The combination therapy not onlyinhibites apoptosis of ischemic skeletal muscle cells partiallyvia downregulation of Bax and upregulation of Bcl-2,but also activates Akt phosphorylation significantly. Theseefficacies may be mediated by the angiogenic potency ofsimvastatin, EPCs, and by the beneficial effects of simvastatinon transplanted EPCs as well.
