- 中文摘要: Tulahuen株枯西氏錐蟲在15隻C3H/Anf品系初生小白鼠作單株分離,其中10隻在第12天在血液內出現寄生蟲,而所有小白鼠在19天內死亡。經體外培養之篩選,二寄生蟲株得以建立,其餘各株則因無法適應於液態終環期培養液中之生長而終被放棄。此二寄生蟲株之體外培養生長曲線及側鞭型變態為終環期錐型蟲體之變態率均於本研究中測知,其對哺乳類動物細胞之感染力雖經體外培養仍得以保持,並亦保有對動物腦、心、肺、食道、胃、大腸、肝、胰、脾、淋巴結、腎、膀胱及肌肉等不同組織之親嗜能力。利用此二單株分離之枯西氏錐蟲作研究,能得到可重覆可信賴之實驗結果。
- 英文摘要: The Tulahuen strain of Trypanosoma cruzi was cloned in 15 C3H/Anf neonatal mice. Ten of these15 neonates became parasitemic before the 12th day and died before the 19th day after the inoculation ofa single bloodstream trypomastigote. Two clones were selected and maintained,while the other isolateswhich did not grow in a liquid metacyclic stage culture (LMC) medium were eventually discarded. Thekinetics of in vitro growth and transformation from epimastigote to metacyclic trypomastigote of thesetwo clones were characterized in LMC medium at 27℃. Infectivities for vertebrate cells in vitro were re-tained by these two clones during the period of cultivation. The tropism for brain, heart, lungs, esophagus,stomach, large intestine, liver, pancreas, spleen, lymph nodes, kidneys, bladder, and skeletal muscles wasalso examined in the mice. The communication describes the establishment and characterization of T. cruzi clones. The utiliza-tion of these cloned parasites should produce some advantages in generating reproducible data in investiga-tions.
