睪丸素抗體血清之製作

以睪丸素用有機合成方法將化物(O-Carboxymethyl) hydoxylamine接於睪丸素之第A環之三位置,因而使之含有自由之-COOH根,利用此根與牛血清白蛋白之ε-胺基酸相接,由此使睪丸素具抗原之作用。將此等抗原注入家兔引發抗體,於抗體血清獲得後,使之與H -睪丸素及睪丸素在緩衝液中作用,並以Dextran-Coated Charcoal分離其結合與自由型式之睪丸素,完成放射免疫法標準曲線之建立。在有機合成之各步驟,以紫外光譜、紅外光譜、薄層分析、Dini-trophenalation等技術,分段檢定各有機合成之物質。根據上述技術可以證明合成物之可信性,並測知與一牛血清白蛋白分子上,至少有三十個睪丸素之分子。以放射免疫測定法得知抗體血清至少可測50 pg以上之絕對量。

Antiserum for testosterone has been inducedin rabbits by using protein conjugates of testo-sterone-3-(O-carboxymethyl) oxime with bovineserum albumin (BSA). By employing solid-phaseabsorption technique, standard curves are esta-blished for radioimmunoassay (RIA) of tes-tosterone in the tissue fluid by using the an-tiserum. With a keto group at the C-3 position,on testosterone reacting with (O-carboxymethyl)hydroxylamine, an oxime as well as a carboxylicacid are formed. Through the mixed anhydrideprocedure, these derivatives couple withlysinesof BSA,Testosterone-3-BSA(T-3-BSA)areformed. After each step of organic syntheses, theproducts have been inspected by ultraviolet, infra-red spectrum and thin layer chromotography toinsure the chemical characteristics. Ultravioletand dinitrophenylation analyses have beenemployed to determine the number of steriodderivatives on one molecule of BSA. There are no less than 30 steriod derivativeson one molecule of BSA. RIA standard curveindicates that the minimum dose of testosteronecan be detected is 50 pg.
