- 英文摘要: Apoptosis has been implicated recently as a prominentresponse of the brain to a variety of insults, such as isch-emia and trauma. In this study, we demonstrate thatapoptosis is a prominent part of the brain's response to athermal insult. To examine the brain's response to a ther-mal insult, a new model of thermal brain injury in thelaboratory rat was developed. Water heated to 60'C waspassed over an area of thinned calvarium for 1 min. Thisresulted in an actual brain temperature of 47-48'C. Auniform area of 2,3,5-triphenyl-tetrazolium chloride pal-for was demonstrated and pyknotic neurons were seenin the area of injury by hematoxylin-eosin staining.Apoptosis was demonstrated by the characteristic DNAfragmentation seen by agarose gel electrophoresis,ApopTag in situ staining and electron microscopy. Thefindings of apoptosis were localized to the area of ther-mal injury and were time dependent, starting 6 h afterthe insult and peaking approximately 18 h after theinsult. This represents one of the first demonstrationsthat apoptosis occurs in the brain in response to a ther-mal injury.
