人參根細胞懸浮培養

鬆軟的人參根癒合組織可作為材料,以建立液態懸浮培養。經過預備實驗後,選用最好的配方作為培養液,含有Murashige-Skoog無機鹽,White有機配方,myo-inositol 100 mg/,蔗糖30 g/,2, 4-dichlorophenoxyacetic acid (2, 4-D) 2 mg/,kinetin 1 mg/,和N-γ,γ-dimethylallylaminopurine (2iP) 1 mg/。無菌空氣通氣攪動培養效果比靜置培養或震盪培養等效果好,細胞增殖迅速且較不聚集。部份細胞分裂與一些藻類細胞分裂方法相似,子細胞形成後原生質呈球形,其外有新生細胞壁,此子細胞與母細胞壁接觸很少,極易被攪動所分離,而形成單細胞個體。

Friable root callus tissues of Panax ginsengC. A. Meyer were used as starting material forthe establishing of batch suspension culture.The medium used for suspension culture, asselected in comparative studies for the bestcallus proliferation in agar-gelled media, con-tained Murashige-Skoog salts, White's organics,myo-insitol 100 mg/, sucrose 30 g/, 2, 4-dichlo-rophenoxyacetic acid (2,4-D) 2 mg/, kinetin1 mg/ and N-γ, γ-dimethylallyaminopurine(2iP) 1 m/. A simple conical glass V-fermenterfor growing root callus cells in suspensionculture was described; it could be used for batchculture or continous culture with further modi-fication to produce large volume of biomass.Aeration and agitation were by continuous bub-bling of germ-free air into the culture. Algae-type cell division has been observed in the batchculture. Namely, the daughter protoplasts mightsecrete about themselves a complete daughtercell wall inside the original mother cell wall.The newly-formed daughter cells expanded, rup-tured the mother cell wall and escaped.
