- 中文摘要: 為提高省產酵母菌體的經濟價值及開拓高溫菌所分祕酵素的應用範圍,本研究從本省各地之土壤及堆肥樣品中,篩選出一株能生產酵母細胞壁溶解酵素之高溫性真菌,此菌株經菌學性質研究之結果,命名為Chaetomium sp. No.25。本菌株生產酵母細胞壁溶解酵素之最適培養基組成為石油酵母0.5%,糖蜜酵母0.5%,酵母抽出液0.1,MgS04 " 7H200.05%,K,HP04 0.05%,KHZPO4 0.05y,,CaCO3 0.1%,。最適培養條件為最初pH值6.0;攪拌速率300rpm二通氣量2:1 ( vol/vol/min ):溫度450C。在上述條件下培養12--.15小時後可獲得最大之酵素活性。本酵素必須在含酵母菌體為誘導物之培養液中才會生產,顯示本酵素屬於誘導酵素。本酵素對省產糖蜜酵母及石油酵母均有溶菌作用,其最適溶菌pH為8:5,最適溶菌溫度為600C。木酵素是屬於以蛋白質水解酵素為主要溶菌作用之酵母細胞壁溶解酵素。
- 英文摘要: In view point of promoting the economic value of local Torula yeastand Petro yeast, and developing the utilized field of thermophilic enzymes,a thermophilic fungus which could produce the yeast cell wall lytic enzymeshas been isolated from the local compost. The taxonomical characte-ristics of this thermophilic fungus were examined and was identified asChaetomium sp. No. 25. The optimal cultural conditions for the productionof yeast cell wall lytic enzyme are as follows,(1)Optimal medium, Petroyeast 0.5%,Torula yeast 0.5%, yeast extract 0.1%,MgS04·7H20 0.05,KHZP04 0.05%,K,HP04 0.05%,CaC03 0.U/1.(2) Optimal fermentationconditions, initial pH 6.0; rate of agitation 300 rpm;rate of aeration 2:1(vol/volymin);temperaturt 450C. After the newly isolated strain wascultivated under the optimal conditions for 12-15 hr, the maximum enzymeactivity was obtained. The yeast cell wall lytic enzyme produced by thisnewly isolated strain belongs to an inducible enzyme. The optimal pHof the yeast cell wall lytic enzyme for the hydrolysis of local Torulayeast and Petro yeast was 8.5 and the optimal temperature was 600C. It wasconcluded that protease played an important role in the lytic action of theyeast cell wall lytic enzyme·
