- 中文摘要: 將自污水中分離之通性菌Bacillus polymyxa D-21以kappa- carrageenan固定做成固定化生長菌粒後,與氧電極配合而組成一密閉反應器式生化需氧量感測器。此感測器能將標準人造廢水(含有等量葡萄糖及麩胺酸之0.1M磷酸緩衝液)中之溶氧迅速消耗,其溶氧消耗速率與人造廢水濃度為10 mg/1或BOD為15 ppm以下時有很好之直線關係(相關係數為0.9919),可供為測定污水中生化需氧量之用途。利用此感測器測定一個樣品需時不到15分鐘。將固定化菌粒於室溫(25℃)下儲放於0.1M磷酸緩衝液(pH 6.0)中,至少可儲存二個月。此感測器之再現性比傳統測定方法佳,其平均相對誤差為5.89%。此感測器具有下述優點(1)操作簡單、快速。(2)穩定性與再現性良好。(3)製備方法簡單、容易、而且便宜。
- 英文摘要: A closed, reactor-type sensor system for rapid estimation of BOD by the use of immobilized growingwhole cells of a facultative bacterium, Bacillus polymyxa D-21, in kappa-carrageenan and an oxygen elec-trode is described. This system consists of a transformer, a recorder, a thermostated water bath (30 ± 1℃),a magnetic stirrer (200 rpm), an oxygen electrode, and a flask containing 10 g (wet weight) immobilizedcell beads of 2-3 mm in diameter. The total time required for an assay is less than 15 min. The calibrationcurves obtained show a linear relationship between the rate of oxygen consumption and the concentrationof standard wastewater solution which contains equal amounts of glucose and L-glutamic acid each at aconcentration below 10 mg/1. The measurement is independent of the pH of the wastewater solutionunaer investigation. The rate of oxygen consumption was reproducible with an average relative error of5.89% when a standard wastewater solution containing 12 mg/1 of glucose and 12 mg/1 of L-glutamicacid was assayed 54 times. When the immobilized cell beads were suspended in a 0.1 M phosphate buffer(pH 6.0) and stored at 25℃, the response of the sensor system remained unchanged for about 30 days.However, based on the observed linear relationship between the rate of oxygen consumption and the con-centration of standard wastewater solution, the sensor system should remain usable for at least 2 months.There is a good correlation between BOD determined by a conventional method and BOD determinedby the proposed method when wastewater samples from a local fermentation factory were assayed.
