腫瘤之粒子X光(PIXE)分析

我們報告幾種腫瘤吸收釔的情況,注射少量硝酸釔於已有腫瘤的裸鼠後,取出肝及腫瘤組織製成靶,由2 MeV質子轟擊而分析碰撞靶所誘發的X光。比較各種腫瘤的含釔量,發現腫瘤吸收釔的含量比健康正常組織的含釔量多4~6倍。為驗證釔是否被腫瘤細胞吸收起見,我們加釔於Raji細胞,然後製成細鮑密度不同的靶,測定各靶中之釔含量,並得釔含量與細胞密度成正比的結果。 此外我們用粒子X光定量分析少量中藥昭示粒子X光分析的有效性。

We investigated the uptake of yttrium to several tumors. Small amount of yttrium nitrate wasinjected into nude mice with several kinds of tumors. Samples of liver and tumors taken out from thesemice were bombarded by 2 MeV protons and the proton induced X-ray emission (PIXE) was analysed bya Ge detector to determine relative concentrations of yttrium taken up by these tissues. We found that theuptakes by these tumors were 4~6 times more than those by normal and healthy controls. In order tolocate where yttrium was retained in cells or otherwise, we added a small amount of yttrium into thecultivated tumor cells for certain time. Then we concentrated the density of a specific cell, Raji cell (aBurkitt lymphoma cell line) in this report, to make the targets of different cell densities for the analysis.We plotted the uptake of yttrium by Raji cell versus cell density and from the slope of the plot we decidedthat yttrium was indeed absorbed by the cell. A small amount of Chinese herb medicine was quantitativelyanalysed to demonstrate the effectiveness of PIXE.
