卵巢激素對黃麴毒素B1肝臟毒性之影響

體重 100 - 120 克史陶氏(Sprague - Dawley)公鼠經腹腔注射黃麴毒素(1 ug/lgaflatoxin ),一小時左右,其肝中總核醣體(total ribosomes)的蛋白質合成活性增高,之後與游離核醣體(free ribosomes)的相似,彼等之蛋白質合成活性漸被抑制。前者至毒素注射後十六小時,後者至廿四小時左右,其活性降至最低,之後又漸增高。經選擇研究此毒素注射後十六小時對體重 100 - 120 克去卵巢白鼠之影響,結果顯示此毒素對它們之影響不若公鼠大,經二週卵巢之腹腔注射(每週三次,每次 1 ug estradiol - 17β/100 g)後,無論給予黃麴毒素與否,均可使游離的與總核醣體之蛋白質合成活性增高,經此卵巢激素處理之老鼠,雖然黃麴毒素仍抑制其游離核醣體之蛋白質合成活性,但對其總核醣體者則反有明顯的刺激作用其間可能機轉曾於本文中加以討論。

Intact male and ovariectomized femaleSprague-Dawley rats of 100 - 120 g bodyweight were injected intraperitoneally withaflatoxin , 1 ug∕g body weight. In themale, their protein synthetic activity oftotal ribosomes was significantly increased1 h after the injection. The activities ofboth free and total ribosomes were thengradually decreased until a maximum inhi-bition was reached at around 16 h afterinjection for total ribosomes and 24 h, forfree ribosomes. The protein synthetic acti-vity of the female rats was determined 16h after the toxin injection. They showedless sensitive to the inhibitory action ofthe toxin than the male. Estradiol - 17βtreatment (1 ug/ 100g body weight, i.p. 3times a week for 2 weeks) stimulatedthe female their protein synthetic activitiesof both free and total ribosomes at 0 or 16h after aflatoxin injection. After thefemale were so treated with estrogen, al-though the toxin injection inhibited theactivity of the free ribosomes, that ofthe total ribosomes was stimulated.
