• 中文摘要： 為提高實際進行酪蛋白水解程序之效率,本研究首先探討胰蛋白(trypsin)固定化以獲得高水解率之方法。以幾丁聚醣為擔體之共價鍵結方式在酵素固定量、酵素活性存活量,重複使用性諸特性均優於物理吸附法。據此將共價鍵結之固定化胰蛋白以固定床式反應器進行連續水解反應,應用回應曲面法,尋找出反應之最佳條件為X/sub 1/(回流率)=0.24,X/sub 2/(基質濃度)=2.28%(w/v),X/sub 3/(pH)=8.28,X/sub 4/(溫度)=37.7.degree.C,以此操作條件,回應曲面法所求得之水解率為9.66%,已接近理論值10.6%,以此條件進行酪蛋白之水解實驗,獲得9.3%之水解率,此結果可提供實際生產酪蛋白磷胜□之參考。
• 英文摘要： With an aim of improving the hydrolysis of Ca- caseinate in a practice, the immobilization method of trypsin tailored to the tryptic hydrolysis to obtain the caseinophosphopeptide was investigated. Covalent binding method with chitosan as a carrier was found to be superior to the physical adsorption method in terms of enzyme loading, relative activity of enzyme remained and reusability of enzyme . A packed-bed reactor, 0.01m in inside diameter, filled with immobilized trypsin, 0.08m in height, was devised to investigate the optimum operational conditions for hydrolysis of Ca-caseinate by employing the Response Surface Method. The optimum operational conditions searched were X/sub 1/ (recycle ratio)=0.24, X/sub 2/(substrate concentration)=2.28% (w/v), X/sub 3/ (pH)=8.28 and X/sub 4/ ( temperature)=37.7.degree.C. Under these conditions, the degree of hydrolysis (D.H.) was calculated to be 9.66% which is close to the theoretical value, 10.6%. Experimentation referring to these conditions resulted in the 9.3% of D.H.
