- 中文摘要: 為提高實際進行酪蛋白水解程序之效率,本研究首先探討胰蛋白(trypsin)固定化以獲得高水解率之方法。以幾丁聚醣為擔體之共價鍵結方式在酵素固定量、酵素活性存活量,重複使用性諸特性均優於物理吸附法。據此將共價鍵結之固定化胰蛋白以固定床式反應器進行連續水解反應,應用回應曲面法,尋找出反應之最佳條件為X/sub 1/(回流率)=0.24,X/sub 2/(基質濃度)=2.28%(w/v),X/sub 3/(pH)=8.28,X/sub 4/(溫度)=37.7.degree.C,以此操作條件,回應曲面法所求得之水解率為9.66%,已接近理論值10.6%,以此條件進行酪蛋白之水解實驗,獲得9.3%之水解率,此結果可提供實際生產酪蛋白磷胜□之參考。
- 英文摘要: With an aim of improving the hydrolysis of Ca- caseinate in a practice, the immobilization method of trypsin tailored to the tryptic hydrolysis to obtain the caseinophosphopeptide was investigated. Covalent binding method with chitosan as a carrier was found to be superior to the physical adsorption method in terms of enzyme loading, relative activity of enzyme remained and reusability of enzyme . A packed-bed reactor, 0.01m in inside diameter, filled with immobilized trypsin, 0.08m in height, was devised to investigate the optimum operational conditions for hydrolysis of Ca-caseinate by employing the Response Surface Method. The optimum operational conditions searched were X/sub 1/ (recycle ratio)=0.24, X/sub 2/(substrate concentration)=2.28% (w/v), X/sub 3/ (pH)=8.28 and X/sub 4/ ( temperature)=37.7.degree.C. Under these conditions, the degree of hydrolysis (D.H.) was calculated to be 9.66% which is close to the theoretical value, 10.6%. Experimentation referring to these conditions resulted in the 9.3% of D.H.
