- 中文摘要: 吾等曾用放射免疫分析法(RIA)測知,Cyanoketone(CK)對蝌蚪離體卵巢分泌雌性素具有抑制作用。此乃因 CK 抑制-3β-HSD 活性所致。在上述實驗中,預期 CK 濃度在 0.1μg/ml時可達 100% 抑制效果,但實際上僅 85%。推其原因,可能有少量雌性素在 CK 處理前合成,存留於卵巢內所使然。故今設計此先期培養(Preincubation)實驗。將卵巢預先在含有不同劑量之 CK 培養中,0,0.001,0.01,0.1, 1 或 10μg/ml,培養30分鐘,再換入同樣之 CK 系列培養液內,繼續培養6小時,收集之,用作 RIA 實驗,測量雌性素之分泌量。結果顯示,CK 之抑制效果在 0.1μg/ml時增加至 95%。由此可知,先期培養,確為測量卵巢分泌量之重要一環。
- 英文摘要: In a previous RIA study we found that cyanoketone (CK) inhibited ovarian secretion of tadpolesin vitro and that this inhibition ettect was through inactivation of -3β-HSD activity. A complete 100%inhibition was expected at a CK dosage of 0.1 μg/ml of the medium, but, instead, it was 85%. The dis-crepancy might be due to the fact that the previous experiments did not preincubate the ovaries with CKin order to get rid of the residual . To this end, the present study was designed. Tadpole ovaries ofRana catesbeiana were preincubated with CK of a dosage of 0, 0.001, 0.01, 0.1, 1, or 10 μg/ml of theKRbb medium for 30 min. The media were discarded. Fresh media with the same series of CK doseswere added to the ovaries and were incubated again for 6 hr. The media were collected for RIA of estro-gen. The results showed the same tendency of estrogen inhibition as the previous study. However, amaximal inhibition effect of 95% was obtained at the dose of 0.1 μg/ml. Therefore, the difference betweennon-preincubation of the previous experiments and preincubation of the present study does exist as wepredicted.
