- 英文摘要: The use of cloud point extraction system with nonionic polyoxyethylene surfactants (C/sub 10/E/sub 5/, C/sub 12/E/sub 5/, and C/sub 14/E/sub 6/) was carried out for direct solubilization and extraction of membrane bound cholesterol esterase (cholesterol ester hydrolase; CEH) from Pseudomonas fl. The effects of various operational parameters (surfactant weight percentage, temperature, phosphate concentration and composition of surfactant mixture) on the CEH partition behavior have been investigated. The partition coefficient and yield changed insignificantly with increasing surfactant (w/v) percentage for both C/sub 12/E/sub 5/ and C/sub 14/E/sub 6/. The cloud point also shows a linear dependence on weight percentage of the shorter headgroup surfactant. The dependence is described approximately as T/sub CP/ = 20.75-0.309W and T/sub CP/= 32.82-0.243W, for C/sub 12/E/sub 4//C/sub 12/E/sub 5/ and C/sub 14/E/sub 5//C/sub 14/E/sub 6/, respectively. After a two-stage separation, an initial 5-fold concentration with 77% yield has been achieved under the condition of 1% (w/v) of C/sub 12/E/sub 5/ at 22.8.degree.C without potassium phosphate addition.
