- 中文摘要: 具有抗癌作用的雞母珠毒蛋白(Abrin)溶於8M尿素,Tris緩衝液pH8.6中,加入2% MSH還原12小時後,可用Sepharose-4B親合性柱體將其A,B二次單元體分開;並由SDS一凝膠電冰可證明二者均已純化 因B次單元體與Sepharose-4B柱體有親台性,故可見B次單元體負責使毒蛋白附著於細胞膜上含有半乳糖構造的接受器(receptor)上,而A次單元體負責抑制細胞的活性。
- 英文摘要: Abrin, an antitumor phytotoxin, was reduced with 2% MSH, O.1M Trisbuffer PH 8.6, in the presence of 8M urea, to split its two subunits. Thesubunits were then separated by chromatography on a Sepharose-4B affinitycolumn, and the isolated A and B subunits were proved to be homogeneousby SDS-gel electrophoresis. The B subunit which could interact withSepharose-4B affinity column, may concern with the ability of toxin tobind to the receptors of cell membrane; while the A subunit which doesnot possess the binding ability, may enter the cell and exert the toxicfunction.
