最近製劑學技術的發展

製劑學技術的發展程序,由經驗期到精密的定量分析期、生物藥劑學及藥物速度論發展期,進入目前的藥物體內配達系統發展研究期。在有效性、安全性的大前題下,依所需目的而發展出各種製劑加工技術及各型製劑。諸如:(1)腸溶性製劑-腸溶性錠劑、腸溶性顆粒、發泡性腸溶錠、腸溶性微粒膠囊。(2)持效性製劑-各種持效、持續性製劑及持效控制特殊裝置。(3)微粒製劑-微粒膠囊、超微粒膠囊、微粒小球、人工脂質膜。(4)化學修飾製劑-前驅藥物、高分子藥物。(5)包嵌化合物。 藥物療法也配合製劑技術的發展,由多次投藥的一般劑型到減少投藥次數的持效性製劑,再進入防止副作用發生,而直接投藥到病害的目的組織或器官的定時、定位、定量的藥物體內配達及控制放出系統。

The process of historical development ofpharmaceutical technology and objects or me-thods of pharmaceutical formulation was de-scribed respectively. Under good stability, avail-ability and safty condition, many new approachsof controlled drug delivery system were devel-oped. Such as enteric-coated preparations, sus-tained release preparations, controlled deliverydevice, microcapsule, nanocapsule, microsphere,liposome, prodrug, polymeric drug and inclusioncompound were discussed in detail. Futuredevelopment of pharmaceutical technology isfocusing on controlled drug delivery systemwhich is defined as drug released from prepara-tions at preprogrammed time, on fixed targetand with definite dose.
