如何改善電顯鏡相片的品質

作者等用了五種底片,以稻熱病菌的同一超薄切片樣品在蔡司109型體外攝影體系,用不同曝光量使底片曝光,再各以最合適的條件沖洗底片以求何者最為合乎電顯鏡攝影之用。結果顯示柯達 2415 得正常反差、愛克發 ortho 25 次之,而富士 Neopan SS 的反差不足。另兩種底片,柯達 EM 4489 及富士 EM FG 雖曾予以較多曝光量均仍全未曝光。 次又用柯達 2415 及愛克發 ortho 25,各以不同曝光量曝光,不同顯影液、不同濃度、不同顯影時間沖洗。結果為減少曝光量、增加顯影液濃度可提高底片的反差;反之,增加曝光量、稀釋顯影液、增加顯影時間可減低底片的反差。如此,工作者可循此原則,於觀察樣品的反差不足或偶或過高時,調整電顯鏡攝影所得底片的反差至正常程度,且由此種底片印洗得品質上乘的相片。

Five kinds of film were used to detectwhich one would be the best for electronmicroscope photographs. The results showedthat Kodak 2415 revealed its normal contrastafter appropriate exposure and development,than it followed by Agfa ortho 25. Thecontrast of Fuji neopan SS was poor. Theother two films, Kodak EM 4489 and FujiEM FG, were not exposed at all at thepreset high density for TFP system of Zeiss109. Further experiments on 2415 and ortho25 films found that less exposure and higherconcentration of developer raised the contrastwhile more exposure, higher dilution of dev-eloper and longer period of developmentdecreased the contrast. This principle couldbe applied to adjust the contrast of the EMnegatives to normal no matter the contrastof the sample is too low or too high. Thehigh quality of EM photographs wouldundoubtedly in turn, be produced from thenegatives with normal contrast.
