- 中文摘要: 嚼食檳榔與口腔癌、口腔黏膜下纖維化症之發生有密切的關係。而檳榔子中的檳榔素(Arecoline)則被認為是檳榔嚼塊中重要的致病因子之一。為了進一步了解檳榔素在嚼食檳榔相關口腔黏膜疾病所扮演之角色,我們以培養之口腔黏膜下纖維母細胞來探討綑胞密度(cell density)、血清濃度(serum concentration)、藥物曝露時問(incubation time)對檳榔素細胞毒性之影響。在濃度低於0.2 mM時,即使以檳榔素處理細胞達六天,細胞毒性並不明顯。但是當培養液中血清濃度為10%時,則以0.2 mM檳榔素處理細胞三天,細胞毒性與細胞制性(Cytostatic)便會表現出來。曝露低密度(1×10 cells/well)之細胞於0.2 mM檳榔素與低血清狀況下三天與六天,分別會導致20%與23%細胞數目的減少。然而,曝露低密度之細胞(1×10 cells/well)於0.2 mM檳榔素與高血清狀況下三天與六天,對細胞數目之影響更大,分別會導致38%與53%細胞數目的減少。另一方面,曝露較高密度之細胞(5X10 和 1×10 cells/well) 於0.2 mM檳榔素與高血清(10%)狀況下六天,對細胞數目之影響則較細胞密度低時為小,分別只導致38%與21%細胞數目的減少。我們的結果顯示,即使在不同的實驗條件下,檳榔素並役有刺激口腔黏膜下纖維母細胞生長之能力。而檳榔素對口腔黏膜下纖維母細胞之細胞毒性則受到細胞密度、培養液中血清濃度以及藥物曝露時間之影響。檳榔素之毒性反應可能和嚼食檳榔相關口腔黏膜疾病的發生有關。
- 英文摘要: Betel quid (BQ) chewing shows strong correlation to the incidence of oral submucous fibrosis andoral cancer in Taiwan. Arecoline, the main areca alkaloid, is considered to be one of the etiologic factors inBQ. To elucidate the role(s) of arecoline in the pathogenesis of BQ chewing related oral mucosal lesions, weused oral mucosal fibroblasts to study the effects of serum concentration, cell density, and incubation timeon the cytotoxic response to arecoline. At a concentration less than 0.2 mM, arecoline was not cytotoxic tooral mucosal cells after 1, 3, and 6 days of incubation. After 3 days of incubation, the cytotoxic and cytostaticeffects of arecoline became evident when the cells were exposed to higher concentrations of arecoline (0.2mM) and serum (10% FCS). Exposure of cells (1 x 10 cells/well) to 0.2 mM of arecoline in 0.5% FCS for3 and 6 days led to a 20% and 23% decrease, respectively, in the cell number, whereas exposure of cells(1 x 10 cells/well) to 0.2 mM arecoline in 10% FCS led to a 38% and 53% decrease, respectively, in cellnumber. At a higher cell density (5 x 10 and 1 x 10 cells/well), 0.2 mM arecoline led to less cytotoxicity(38% and 21% of decreasing in cell number, respectively) after 6 days of incubation. Our results indicatedthat arecoline was not mitogenic to oral mucosal fibroblasts, and that the cytotoxic and cytostatic effects ofarecoline on oral mucosal fibroblasts could be modulated by the changes in the cell density, serum con-centrations, and incubation time.
