單向免疫擴散法檢定家蠶核多角體病毒感染之研究

以鹼溶液溶解之家蠶核多角體注射兔子經 5 週後可得力價為 256 對抗病毒之抗血清。經雙向擴散法確認此抗血清只和病毒及病蠶起反應。進一步分析,得知此抗血清含有對抗病毒顆粒及對抗多角體蛋白質的成分存在,可供早期檢定病毒之用。 用單向免疫擴散法測知含 2%之抗血清為反應最適濃度,並測定在定性場合下,25°C 為最適宜的反應溫度,且2小時內即可產生明顯的沉澱環以資檢定病毒。 實際操作時,將含有 2%抗血清的瓊脂 4 ml 倒入內徑 8.5 cm 的培養皿內,每隔約 1 cm的距離打出直徑 1 mm 的反應洞,每一擴散盤可打洞 30~50 個。將受檢蠶以鹼溶液研磨 20分鐘後,滴加 0.4~0.8μ1 的研磨液至反應洞內,在 25°C 讓其反應2小時,即可計算沉澱環產生的數目。測定結果,初孵化一齡蠶第二天及二、三齡蠶第三天即可檢出潛伏病毒,接種後96小時第一至第三齡罹病蠶檢出率達 70~100%。 本法發展特異性高,簡便,快速及經濟之家蠶病毒檢定系統,可供調查蠶病及防疫之應用。

An antiserum against nuclear polyhedrosisvirus (NPV) of the silkworm, Bombyx mori, wasobtained by injecting the alkaline-dissolved poly-hedra into rabbit for over 5 weeks, its titerbeing ca. 256. The antiserum could react withpurified NPV and extracts of NPV-infectedsilkworm to form precipitated bands by usingdouble immunodiffusion. Further studies foundthat this rabbit antiserum contained both anti-virion and anti-polyhedrin components, therefore,it is useful for detection of the early infectionby NPV. Single radial immunodiffusion showed thatthe antiserum at 2% was best for reacting withantigens, its optimum reacting temperature being25°C. Rapid detection of NPV infection maybe obtained within 2 hours in terms of ringformation. For practical implementation, single radialimmunodiffusion was conducted as follows. Ona 8.5-cm petri dish layered a medium of 2%antiserum in 0.8% Difco noble agar dissolvedin boiling phosphate buffer, pH 7.0, 30-50 wellsof 1 mm in diameter being set apart from eachother by 1 cm were punched out in each agarplate. The insect homogenates to be tested werefilled into each well at0.4-0.8 μl using a capillarytube. The plate was incubated at 25°C for 2hours, and then the precipitated rings wereformed and visible. Larvae with latent NPVcould be detected 2-3 days after inoculation, thedetecting rate 96 hr post-inoculation in 1st-3rdinstar being 70-100%. This serological method is specific, effective,simple and economical for detecting the NPVinfection in the silkworm, and is also useful forsurvey on NPV occurrence in the sericulturalfarms.
