- 英文摘要: In a combination of biochemical and immunoelectron-microscopical approaches we studied intracellular traf-ficking and localization of the endoplasmic-reticulum(ER)-formed complexes of murine MHC class II moleculeI-A and an antigenic peptide Eα52-68 covalently linkedto its β-chain. The association with the peptide in the ERleads to sharp acceleration of the intracellular traffickingof the complexes to the plasma membrane. Within thecells, Eα52-68:1-A complexes accumulate in the multive-sicular MHC class II compartment(MIIC), but not in dens-er multilaminar or intermediate type MIICs. The changesin the trafficking of ER-formed complexes result solelyfrom the presence of the tethered peptide, since wild-type class II molecules traffic similarly in bare lympho-cyte syndrome cells and in wild-type antigen-presentingcells.
