- 中文摘要: 目前世界各國通用之木質素定量法,大抵衍生自美國製漿造紙技術協會之標準法,其操作過程中,以第二次水解最為費時,此亦為各方研究改進之焦點。 於木質素存在下之水解物中,定量其碳水化合物,常受干擾,最近經Willis等氏以特定呈色反應獲得滿意結果,本研究應用其方法,來選擇費時短且劣解少之第二次水解條件。 測定木材中木質素的改良條件為:第一次水解——每克木材(絕乾重)加入72%硫酸溶液15 ml ,溫度維持於20±1℃下,時加攪拌2小時;第二次水解——上述水解物加水稀釋成3%硫酸濃度,放入高壓釜中,於溫度121℃與壓力103 kPa下,水解30分鐘。
- 英文摘要: The current standards used for quantitative determination of lignin in wood are more or less derivedfrom the TAPPI standard T 222 os-74 and Useful Method 250. But their analytical procedures are timeconsuming and cumbersome, particularly in the secondary hydrolysis. This procedure has called for moreattention to its modification. Recently, a rapid colorimetric method for determining the total carbohydrate content of hydro-lyzate in the presence of lignin was established by Willis et al.(1985). This method was adopted here inhelping to select suitable conditions in the secondary hydrolysis with minimum time and decomposition. An improved method for the determination of lignin in wood is suggested as follows: Primaryhydrolysis - 15 ml of 72% sulfuric acid was added to each gram of oven-dry wood meal. The mixturewas stirred frequently and kept at 20±1±℃ for 2 hours to assure complete solution. Secondary hydrolysis- The hydrolyzate was diluted to 3% concentration of sulfuric acid and autoclaved under 121℃ and kPafor 30 minutes.
