- 中文摘要: 本報告乃敘述有關鰻魚腎臟細胞株EK-1之建立。本細胞株在Leibovitz's L-15 medium 加10%之牛犢血清之培養基內於溫度32 1°C,經過18月已繼氏培養110次。EK-1 細胞株之細胞形態包括含大核及小核之上皮細胞以及纖維性細胞。於此細胞株中亦可見少許的多核上皮細胞。將 100 個分裂中期之EK-1 細胞做染色體分析時,其結果顯示染色體數目之分佈範圍由21至62並形成雙向分佈,而以38個染色體之細胞為最多。於低濃度細胞培養時則發現EK-1細胞之表面附著效應(Plating Efficiency)甚低。本實驗亦證實EK一1 細胞株細胞株可接受EVE, EVA及EVEX等三種病毒之感染,而無細菌、真菌及微漿體之污染。
- 英文摘要: A permanent cell line designated EK-1 was developed from Japanese eel (Anguilla japonica) kidney.The cell line has been passed for 110 transfers in a period of 18 months in Leibovitz's L-15 medium plus10% foetal calf serum at a temperature of 32 1°C. The EK-1 cell line consists of epitheloid cells withmicro- or macro-nuclei and fibroblastic cells. Multinucleated epitheloid cells were present in most of thecultures. The results of chromosome analysis for the EK-1 cell line revealed that the chromosome numberscounted from 100 metaphase cells show a bimodal distribution ranging from 21 to 62, with a modal num-ber of 38, which is similar to that of normal eel somatic cells. When subjected to low seeding densities,the EK-1 cells showed a low plating efficiency, which might result from the non-transformation of the cells.It was also shown that the cells were susceptible to Eel Virus America (EVA), Eel Virus Europe (EVE) orEel Virus Europe X (EVEX). The EK-1 cell line was demonstrated to be free of bacterial, fungal andmycoplasmic contamination.
