- 中文摘要: 培養基MCDB 153中若不加入“表皮生長因子”,皮膚角質細胞倍殖期長,生長緩慢,甚至可引起生長停止現象。當加入“表皮生長因子”在培養基中時,則角質細胞的分化作用延遲,同時刺激細胞增生。本結果明顯地指出“表皮生長因子”能有效加強皮膚角質細胞的增生作用,但對分化作用影響較弱。所以“表皮生長因子”在角質細胞培養中為一重要之細胞生長因子。完整性MCDB 153培養基同時可以有效抑制纖維母細胞過度生長,所以不需以NIH/3T3細胞作滋養層,經血清刺激後則可培養出完整可移植之表皮。這些表皮經細胞形態學檢查,染色體分析及裸鼠細胞接種檢查,均為正常。所以可為嚴重灼傷患者提供自體皮膚移植的來源。
- 英文摘要: Epidermal growth factor is an important element in maintaining keratinocyte proliferation andmaturation. To evaluate its effect on keratinocyte growth in vitro, human foreskins were cultured. Theepidermal keratinocyte growth in culture was separated into two stages by a conditional medium: theproliferation stage, in which the cells were maintained in a monolayer; and the differentiation stage, inwhich the cells grew to stratification and keratinization. The keratinocytes were cultured in various con-centrations of epidermal growth factor, and their morphology and growth behavior were closely observed.Our results demonstrated that cultured keratinocytes grew in a confluent layer under the influence ofepidermal growth factor. In constrast, in a culture without epidermal growth factor, the proliferation rateof cultured keratinocytes slowed down and eventually the cells stopped growing. During serum stimula-tion, with or without additional exogenous epidermal growth factor, the cultured keratinocytes grewcontinuously to the normal terminal differentiation. Under this two-stage culture model, the culturedkeratinocytes could grow into an intact sheet of graftable epidermis.
