氯化鎘對小白鼠骨中鹼性和酸性磷酸酯解?之影響

本實驗主要在探討氯化鎘 (Cdcl) 對小白鼠脛骨中鹼性及酸性磷酸酯解? (ALP和ACP) 的活性,以及RNA和蛋
白質的合成等影響。雄性ICR種系的小白鼠每週用CdCl 由腹腔注射處理,其劑量分別為200、300或400 g/100g體重。
注射處理後一週起,每週犧牲十隻以上之小白鼠,如此延續六週。結果顯示:氯化鎘抑制 ALP 的活性,且與劑量成正比
,但對ACP的活性,卻具提升的作用;可溶性磷降低,而可溶性鈣增加;RNA及蛋白質的合成機制受損,脛骨中鎘含
量增加,而鋅含量減少。氯化鎘對ALP、ACP及RNA合成的影響,除了活體 (in viro) 實驗外,在離體 (in vitro) 實
驗更獲得證實。如此,由數據可得知:氯化鎘的處理,使得 ALP 活性下降,而 ACP 活性卻上升,脛骨的鈣化作用可能
被延滯,而加速了脫鈣作用。因此,當動物經長期的鎘處理或暴露時,將會嚴重導致骨骼產生脫鈣作用,並減少骨骼內的
基質。

This research was aimed at investigating the effects of cadmium chloride (CdCl ) on the alkalineand acid phosphatase (ALP and ACP) activities and the RNA and protein syntheses of the mouse tibia.The male mice of ICR strain were injected weekly i.p. with three different doses of CdCl , namely, 200,300 or 400 g/l00g body weight. More than ten mice of each group were sacrificed each week startingone week post-injection and each experiment lasted for six weeks. The results showed that due to cadmiumtreatment, there were dose-dependent reductions of ALP activities. In contrast, the ACP activities showedenhancements. The soluble phosphorus was decreased while the soluble calcium was increased. Thebiosyntheses of RNA and protein were impaired. Cadmium in the tibia was augmented while zinc contentwas diminished. The in vivo results on the ALP, ACP and RNA biosyntheses were confirmed by the in vitroones. Thus, our data indicated that as a result of cadmium treatments, the activity of the ALP was in-hibited, that of the ACP was elevated, and the ossification of the tibia might have been retarded while itsdecalcification accelerated. Consequently, the long-term administration of, or exposure to, cadmium wouldinduce serious decalcification of the bone and diminution of its matrix.
