新培養成功人體肝癌細胞株的超微結構

粒,但有類似正常肝細胞有的溶解體、油滴、膽小管等構造,這些構造在這三個細胞株又有程度上的差異,如Hep GZ
具有和正常肝細胞一樣的膽小管,Hep 3B膽小管構造雖可辨認但不明顯,HA-22T則具有不太可辨認的形態。這些發現
加上異染色質在這三個細胞株的多寡和以前所作生化、免疫等實驗結果吻合,顯示出HA-22T在形態上及功能上具有最
少正常肝細胞的特徵。因此,HA-22T被認為是屬於分化較差的細胞株,但它和完全分化的Hep G2,及部分分化的Hep
3B可提供作為研究人體肝癌生物學上研究的珍貴材料。

A human cell line (HA-22T/VGH) newly established from hepatoma patient was studied compara-tively with two other well-defined hepatoma cell lines (Hep G2 and Hep 3B) by electron microscopy. Theresults showed that these three cell lines did not have round-shaped nuclei, peroxisomes, and glycogen par-ticles which are found in the normal hepatocytes. Lysosomes, Lipid droplets, and bile canaliculus-likestructures were found in these three cell lines, however, with some degrees of similarity and dissimilarity.As to bile canaliculus-like structures and cell junctions, Hep G2 contained well-defined structures as normalhepatocytes; Hep 3B had recognizable structures whereas HA-22T had ill-defined morphology. These find-ings with the results of amounts of heterochromation in the three cell lines were compatible with the pre-vious biochemical and immunological results indicating that HA-22T obtained less morphological and func-tional features of hepatocytes. Therefore, we postulated that HA-22T was a poorly differentiated cell line.Thus HA-22T will be useful and valuable for studying the biology of human hepatoma along with the well-differentiated Hep G2 and partly differentiated Hep 3B.
