- 中文摘要: 本研究探討利用Pseudomonas amyloderamosa突變株JD210生產isoamylase時之營養需求。在三角瓶振盪培養中,最適起始pH值為5.0 Maltose及Soybean protein hydrolyzate為isoamylase生產之最佳碳源及氮源。金屬離子Zn+2與Cu+2對isoamylase生產有嚴重的抑制現象。其他金屬離子、磷酸鹽及界面活性劑等則對酵素生產無顯著之影響。根據auxanography及單一去除實驗之結果顯示,菌體生長需要proline與isoleucine兩種胺基酸。而且,0.1% proline的添加可使酵素產量提高約30%。
- 英文摘要: Nutritional requirements for the production of isoamylase by Pseudomonas amyloderamosa mutantstrain JD210 were investigated. The optimal initial pH for enzyme production in shake-flask cultivationwas 5.0. Maltose and soybean protein hydrolvzate were found to be the best carbon source and nitrogensource, respectively. The enzyme production was drastically inhibited by Zn+2 and Cu+2. Other metalions phosphates and surfactants exhibited no significant inhibitory or accelerating effect on enzyme pro-duction. According to auxanography and single omission experiments, proline and isoleucine were requiredfor growth. The supplement of 0.1% proline increased enzyme production by around 30% compared withno addition.
