海鰻性腺刺激素結構單元體之性質

海鰻性腺刺激素經SDS膠體電泳及 N-末端分析證實由兩個不同的結構單元所組成;兩個單元體可經0.1M 丙酸的處理而解離,解離後可利用 Phenyl-Sepharose CL-4B疏水性交互作用色層分析將之分離。在以鯉魚離體睪丸組織產生雄性素之性腺刺激素檢定實驗裡,單元I不具活性,但單元II則有原來激素活性的2.8%;在放射免疫檢定法實驗裡(利用第二種抗體之分離法),單元I對單元II抗體有1%之反應,而單元II對單元I抗體有6%之反應。單元I及II之氨基酸組成與鯉魚性腺刺激素之相對單元頗類似,但不同於哺乳類的α及β單元之組成。

Pike eel gonadotropin consisted of two non-identical subunits as tested by SDS gel electrophoresisand by N-terminal amino acid analysis. Subunits were able to dissociate in 0.1 M propionic acid and weresuccessfully separated by hydrophobic interaction chromatography on Phenyl Sepharose CL-4B. SubunitI showed no gonadotropic activity and subunit II showed 2.8% activity of the native gonadotropin moleculeas assayed by stimulation of androgen production in carp testis in vitro. Antisera of both subunits wereprepared, and subunit I showed 1% cross reactivity to the antiserum of subunit II while subunit II showed6% cross reactivity to the antiserum of subunit I as demonstrated in their radioimmunoassay by usingsecond antibody method. Amino acid and carbohydrate composition of both subunits were very similar to those of cor-responding subunits of carp gonadotropin but differed from those of cY- and p-subunits of mammaliangonadotropins.
