心臟毒素對大腦、肝臟及癌細胞ATPases之作用

大小與細胞生長有密切關係,因此本研究擬探求鼠腦,肝臟及歐立氏腹水癌細胞之
ATPase, - ATPase 及 - ATPase 之活性,並研究心臟毒素對此三種?之作用,結
果顯示大腦含-ATPase 活性最高,肝臟含-ATPases 活性最高,而-
ATPase 活性在各組織均很低,歐立氏腹水癌細胞的特徵是含此三種?活性特別低,而且對心臟
毒素之反應與大腦及肝臟不同,大腦及肝臟之-ATPase 及-ATPase 皆可為
心臟毒素抑制,而大腦之-ATPase 可為毒素促進,肝臟之-ATPase 則為毒素抑
制,此結果顯示可能這三種 ATPases 所含之磷脂不同,因毒素對 ATPases 之作用主要與磷
脂產生離子反應的結果,本研究發現外加磷脂可拮抗毒素在 ATPases 的作用再度證明此種想
法。而歐立氏腹水癌細胞的三種 ATPases 均不受心臟毒素抑制,反而有促進作用,這些結果
顯示癌細胞膜的磷脂與正常細胞可能不同,因此導致 ATPases 活性特低,也可能因此引起不
正常生長。d

-ATPase, -ATPase and-ATPase activities of cerebral cortexand liver isolated from the mouse as well asthose of Ehrlish ascites tumor cells wereassayed and the effects of cardiotoxin on theseenzymic activities were studied. Among thesethree tissues, -ATPase activity ofthe cerebral cortex was the highest, while-ATPase activity was found to behighest in the liver. -ATPase activityof all tissues was quite low as compared withthe other two ATPases. Ehrlich ascites tumorcells not only contained very low ATPaseactivity but also responded to the effect ofcardiotoxin differently. Cardiotoxin inhibited-ATPase and -ATPase of thebrain and liver, inhibited -ATPase ofliver but activated that of brain, but onlyactivated ATPases of Ehrlich ascites tumorcells. Since the effect of cardiotoxin can bereduced by the exogenous phospholipids,cardiotoxin altered the enzymic activity byinteracting with the phospholipid moiety ofthe cell membrane or the enzyme itself·These ressults suggest that Ehrlich ascitestumor cells have a different phospholipidcompositions of the cell membrane, whichmay regulate the ATPase activity and cationictransport of the membrane.
