- 中文摘要: 利用酸-丙酮萃取法,離子交換層析法與分子過濾法由豬腦提取一種鹼性蛋白質並純化至均質狀態。此蛋自質可引發天竺鼠之實驗性腦脊髓炎(Experimental Allergic Encephalomyelitis),同時具有下列特性生:含大量之甘氨酸與鹼性氨基酸;不其α-螺旋構造;不含胱氨酸或半胱氨酸;含有甲基精氨酸(Methylated Arginine);具有很高等電點(PI>10)。用SDS-膠體電泳法測得分戶量為20,000;而在水溶液中以分子過瀘法測得分子量為36,000顯示有雙元體存在。
- 英文摘要: An encephalitogenic myelin basic protein was isolated from fresh frozen pig brains by acid-acetoneextraction and purified to homogeneity by CM-cellulose column chromatography and gel-filtration on aSephacryl S-200 column. In addition to its ability to induce experimental encephalomyelitis in guinea-pigs,this protein exhibited the characteristic properties of mammalian myelin basic proteins: high content ofglycine and basic amino acid residues; presence of methylated arginine residues and absence of cystine orcysteine; absence of a-helix as revealed by circular dichroic studies in aqueous solution and high isoelectricpoint (pl>10). The molecular weight as determined by SDS-polyacrylamide gel electrophoresis was a-round 20,000, in contrast to 36,000 obtained by gel-filtration on a Sephadex G-100 column. The lattervalue suggested that the protein might exist as dimers in aqueous solution.
