The above-described characteristics of HKB cellsindicate that HKB11 is a favorable cell host for the pro-duction of human therapeutic proteins.Cell fusion techniques were used to derive mammalianhost cell lines suitable for large-scale production of ther-apeutic proteins. Although the 293S cell line, of humanembryonic kidney origin, is an excellent host cell formammalian gene expression, these cells have a tenden-cy to form large and tight aggregates in suspension cul-tures and bioreactors. To solve the problem of aggrega-tion,293S cells were fused to a human suspension cellline, 2138 (a Burkitt's lymphoma derivative), using poly-ethylene glycol (PEG). The PEG-treated 293S and 2138cells were selected in a medium supplemented withhypoxanthine-aminopterin-thymidine and G418 (1 mg/ml) to eliminate nonfused cells. These hybrid clones,designated as HKB (hybrid of kidney and B cells), arenegative for endogenous immunoglobulin expression.Most clones are readily adaptable to serum-free suspen-sion culture under shaking conditions without forminglarge and tight aggregates.
