• 中文摘要： 從飯匙倩毒分離山來的結晶毒素－－Cobrotoxin 分子中含有二個酥氨酸基。欲明瞭酥氨酸基與 Cobrotoxin 之致死毒性與抗原性之關係，以化學修飾法與分光光度滴定法探討其在毒素分子中之狀態。 結晶毒素與 Tetranitromethane 在 0.05 M Tris-HCl buffer（pH 8.0）中反應，所得修飾毒素以氨基酸分析結果，僅一個酥氨酸基被修飾而巳，而其毒性與抗原性不受影響，假如此反應在 5M guanidine-HCl 存在中進行時，則兩個酥氨酸基都被修飾，而毒性與抗原性則完全消失。 以分光光度滴定法測定結果，得知結晶毒素分子中一個酥氨酸基與一般蛋白質分子中正常酥氨酸基一樣，為正常解離，其 pKa＝9.65，而另一個包藏在分子內部者，須在 pH 11.3 以上才能解離。 為進一步決定與生物活性有關之酥氨酸基的位置，將僅一個酥氫酸基被修飾之毒素，經過RCM 化後，以 Trypsin 消化之，以高壓濾紙電氣泳動法以及濾紙色層分析法分離含有 Nitro-tyrosine 之 peptide。經氨基酸分析結果，得知此 peptide 由等 mole 之 Arginine, Glycine以及 Nitrotyrosine 所組成。一方面利用 PTC 法決定此 Tripeptide 之氨基酸排列順序為 Gly-Nitrotyr-Arg。由結晶毒素之第一次構造，則可知此 peptide 為第 34～36 之部份。因此證明不容易被修飾而與生物活性有關者為第25個酥氨酸基。 測定這些修飾毒素之圓偏光二色性結果，得知一個酥氨酸基被修飾者，其 CD spectrum 與原毒素大致相同，但兩個都被修飾時，即原有的 CD spectrum 完全消失。此結果表示結晶毒素分子中之第25個酥氨酸基在維持毒素之生物活性所必頓之立體構迭上佔有極重要之作用。
• 英文摘要： Spectrophotometric titration of cobrotoxin showed that one of the two tyrosylgroups was titrated freely with a normal apparent pK of 9.65 and the remainingone ionized with time only after irreversible conformational change had occurredat pH above 11.3. Reaction of cobrotoxin with tetranitromethane resulted in selectivenitration of the tyrosine 35 without altering the biological activity and conformationof the toxin. However, when the tyrosine 25 which appears to be "buried" in themolecule was modified, the toxin lost its biological activity and conformationalchange occurred concurrently. These findings indicate that the intact tyrosineresidue at position 25 in cobrotoxin is essential for maintenance of the active confi-guration of the toxin.
