- 中文摘要: The stabilities of triplexes with two sequences, A/sub n/ and (AG)/sub n/, were studied by 5'-C/sub 2/ A/sub 16/C/sub 2//5'-T/sub n/ (n=12, 14 and 16) and 5'-(AG)/sub 8//5'-(CT)/sub n/ (n=4, 5, 6, and 7) systems by variation of the chain length (of oligopyrimidine sequences), the composition of base triad(T.bcdot.A.bcdot.T versus C/sup +/.bcdot.G.bcdot.C), pH(acidic and neutral), and the concentration of NaCl and MgCl/sub 2/. No stable C/sup +/.bcdot.G.bcdot.C base triad was formed at neutral pH. However, C/sup +/.bcdot.G.bcdot.C is more stable than T.bcdot.A.bcdot.T in acidic conditions. The triplex formed from the 5'-C/sub 2/A/sub 16/C/sub 2/ and T/sub 12/ mixture with 12 T.bcdot.A.bcdot.T units has a melting temperature Tm 11.degree.C. The triplex formed by 5'-(AG)/sub 8//5'-(CT)/sub 4/ with four T.bcdot.A.bcdot.T and four C/sup +/.bcdot.G.bcdot.C base triads shows Tm 41.degree.C. Tm of 5'-C/sub 2/A/sub 16/C/sub 2/.bcdot. [ T/sub 16/ ] /sub 2/ triplex increased proportionally to the increased NaCl and MgCl/sub 2/ concentrations. Tm of 5'-(AG)/sub 8/.bcdot. [ 5'-(CT)/sub 7/ ] /sub 2/ became constant when NaCl and MgCl/sub 2/ attained 0.5M and 5mM, respectively. The properties of triplexes with A/sub n/ and(AG)/sub n/ sequences were discussed.
