- 英文摘要: Lysosome-associated membrane protein (LAMP)-1, oneof the major protein components of the lysosomal mem-brane, is upregulated in the human glioblastoma celllines, U-373 MG and LN-Z308, which undergo cisplatin-induced apoptosis. These human brain tumor cell linesdemonstrated apoptosis in response to cisplatin/nifedi-pine treatment. Both cell lines demonstrated an apoptot-is response by more than one criterion. Apoptosis wasdemonstrated by DNA fragmentation techniques such asDNA laddering, ApopTag in situ labeling,and an ELISA-based method of detecting liberated oligosomes. Thesecells also had characteristic morphologic changes andupregulation of bax consistent with apoptosis. LAMP-1expression at the protein and mRNA level was examinedand found to increase with cisplatin/nifedipine treat-ment. LAMP-1 expression was examined using indirectimmunofluorescent staining, Northern blot analysis andWestern blot analysis. The finding of an augmentation ofLAMP-1 in these cells induced to die is enigmatic. Thesefindings raise the possibility of LAMP-1 involvement inthe apoptotic process.
