無子西瓜的組織培養莖頂和側芽增殖技術和植珠

三倍體無子西瓜(Citrullus vulgaris, L.)農友新一號十二天大之幼苗莖頂,在含?MS
無機鹽類、White有機配方及2 mg/l benzyladenine (BA)的培養基,可誘發6-8個不定
芽。分割後的不定芽置於含MS無機鹽類,0.4 mg/l thiamine HCI, 1 mg/l kinetin (KA),
1 mg/l gibberellic acid (GA)及0.5 mg/l indoleacetic acid的培養基中,可抽長,發根
形成幼苗。帶有腋芽的切節在含有MS無機鹽類,White有機配方,1 mg/l KA和1 mg/l
GA的培養基上,腋芽形成獨立枝條;在含MS無機鹽類、White有機配方、2 mg/l BA
及1 mg/l GA的培養基上有多量的不定芽形成。

Triploid watermelon (Citrullus vulgaris L.)shoot tips of 5 mm long cultured on an agar(0.7%)-gelled medium contained 1/2 Murashige-Skoog (MS) salts, White's organics and 2 mg/lbenzyladenine (BA), produced maximum adventi-tious bud proliferation at basal portions ofcultured pieces. Then the adventitious budswere excised and cultured on a medium con-taining MS salts, 0.4 mg/1 thiamine HCI, 1 mg/lkinetin (KA), 1 mg/l gibberellic acid (GA) and0.5 mg/l indoleacetic acid. The excised budselongated to approx. 4cm within 4 weeks.Excised stem nodes maintained on a mediumcontaining MS salts, White's organics, 1 mg/lGA and 1 mg/l KA, established as shoots,while cultured on the same basal medium contain-ing 2 mg/l BA instead of 1 mg/l KA, adven-titious bud proliferation resulted. Incubationconditions were 12 hr light of ca. 2 Klx at 25-28°C.
