硫酸鋅在大白鼠中對乙醇引發性胃潰瘍的防止

本實驗為研究硫酸鋅對酒精所產生胃潰瘍的作用機轉。口服硫酸鋅在劑量17.5及35.0 mg/kg能防止乙醇(100%)所造成的胃黏膜潰瘍。它的防止作用為從給藥後30分鐘至24小時,利用phytate餵食所產生缺鋅現象更能加強乙醇性的潰瘍作用。乙醇能減低胃黏膜上的黏液,更能增加在胃腔中組織胺、蛋白質及分泌物的含量,但預給硫酸鋅能改變上述的反應。Carbenoxolone能增加胃黏膜黏液的分泌,同時防止胃潰瘍的發生。Diphenhydramine能明顯地減低乙醇引發性胃潰瘍的產生,但Cimetidine並不產生效果。 故從這些實驗結果觀之,硫酸鋅對防止乙醇所產生的胃潰瘍有下列兩個基本作用機轉:第一是增加胃黏膜的黏液分泌,進而加強胃黏膜的防衛性,這種作用正由減低胃液中的蛋白質含量反應出來。第二是減低組織胺從胃中釋放出來,進而減低其在H 接受器上的病理作用。

The mechanism of action of zinc sulfate on gastric ulceration induced by ethanol was studied. Zincsulfate at the doses of 17.5 and 35.0 mg/kg given orally prevented gastric glandular ulcer produced byabsolute ethanol. The protective action started as early as 30 min and wore off 24 hr after drug administra-tion. Moreover, zinc ion deficiency induced by phytate-feeding aggravated the ulceration. Ethanol signifi-cantly decreased the amount of mucus on the gastric glandular mucosa and increased the total amount ofhistamine, protein and volume of secretion in gastric lumen. Zinc sulfate pretreatment reversed the aboveparameters. Carbenoxolone pretreatment increased the mucus secretion and prevented the ulcer formation.Diphenhydramine but not cimetidine also markedly reduced gastric ulceration induced by ethanol. Thus,from these data, it is suggested that zinc sulfate prevented ethanol-induced gastric ulceration by two basicmechanisms. Firstly, increase of mucus secretion from gastric glandular mucosa stregthened the mucosalbarrier. This was demonstrated by decreased protein leakage into gastric secretion. Secondly, reductionof histamine release from intramucosal stores alleviated the pathological actions of histamine probablythrough H receptors on stomachs.
