Whether the expres-sion of inducible nitric oxide synthase (iNOS) is affectedby exercise is unknown. We therefore investigated theeffects of chronic exercise on iNOS and eNOS expres-sion in isolated rat aortic endothelial and smooth musclecells separately. Five-week-old male Wistar rats wererandomly divided into control and exercise groups. After10 weeks of running training, animals were sacrificedunder ether anesthesia. The standard curve quantitativecompetitive reverse transcriptase-polymerase chainreaction method was used to quantify NOS mRNA ex-pression in isolated endothelial/smooth muscle cells. Toevaluate the functional role of iNOS, we examined phe-nylephrine-induced vascular responses with or with-out pretreatment with aminoguanidine. We found that(1) expression of iNOS and eNOS mRNA in endothelialcells was increased by chronic exercise and (2) chronicexercise blunted phenylephrine-induced vascular re-sponses, probably by increasing NO release via iNOS.Our results show that chronic exercise increases bothiNOS and eNOS gene expression in endothelium.
