- 英文摘要: Opioid drugs such as methadone or buprenorphine areoften used in the management of pregnant addicts.These drugs are generally thought of as nonteratogenicand preferable to repeated cycles of withdrawal in utero.However, evidence exists that perinatal exposure tothese opioids delays and disrupts cholinergic develop-ment, particularly in the striatum. Acetylcholine (ACh)content and the expression of choline acetyltransferaseprotein and mRNA are reduced in the early postnatalperiod by prenatal opioid exposure in the rat. Althoughthese indicators of the cholinergic phenotype return tonormal levels over time, the activity of the cholinergicneurons remains disrupted, with a large increase in AChturnover rate. The mechanism of these effects is un-known, but may involve changes in the expression ofnerve growth factor, which is reduced by opioid expo-sure.
