雄性與雌性激素對鼠體組織中多胺濃度之調節作用

多胺是指生體細胞中廣泛分佈著的腐肉鹼(Putrescine),精胺質(Spermidine)及精液素(Spermine)。此類化合物最近有很多研究。由文獻指出癌細胞與正常細胞的生長速率與多胺之濃度息息相關,更有進者,細胞中DNA,RNA及Protein 之生體合成速率也與多胺之濃度有密切的關連。 本研究利用Sprague-Dawley品系老鼠來作研究,並利用本研究室首創之Dabsylation-HPLC Procedure來定量鼠體中各種組織多胺之含量。若將雄性鼠之睪丸去除,其攝護腺中之多胺有明顯的下降,可注射Testosterone enantha-te以恢復之。若將雌性鼠之卵巢去除,則其子宮中之多胺有明顯的下降作用,可用Estradiol Valerate來挽回此下降趨勢。其他組織如大腸、肺部之多胺濃度亦受此兩種激素之影響,但較不顯著。 在各組織中以精胺質之含量最高,受性激素之控制也最為敏感,同時,它之含量與細胞之成長,或 DNA,RNA 與Protein 之合成也最有影響力。 多胺對各種合成性反應(anabolic response)有密切之關連,但其分子作用機轉,尚未清楚。由各種研究結果顯示多胺可能作為激素刺激基因表現過程中很重要之橋樑。

The levels of putrescine, spermidine and spermine in various tissues of male and female rats (Sprague-Dawley strain) were estimated by a newly developed dabsylation-HPLC procedure. Significant depressingeffects of orchiectomy and ovariectomy on the levels of these amines were demonstrated separately in theprostate of male rats and the uterus of female rats. Exogenous testosterone enanthate and estradiol valeratewere administered intramuscularly to monitor the polyamine levels in the orchiectomized and ovariec-tomized animals. The levels of polyamines in rat prostate and uterus were intimately modulated by the presence oftestis and ovary respectively and this modulation was probably mediated by the circulatory androgenic andestrogenic hormones. Spermidine was found to be the most abundant polyamine in all tissues and itsconcentration was most sensitive to the control of hormonal and growth stimuli. The molecular mechanism of the polyamine intervention in certain anabolic response is discussedand it is suggested that polyamine may act as sequential mediator or activator in triggering the specificgene expression at particular chromosomal loci in response to certain hormone stimulation.
