放射免疫分析法測定黃?毒素B1之含量

本研究乃參照其他實驗室之放射免疫分析法,建立並發展成一可行於臺灣之放射免疫分析測定黃麴毒素B1(AFB1)之系統。黃麴毒素B1經合成為AFB1-1-(o-carboxymethyl)oxime後,分別與BSA或PL結合成免疫原。十二隻經接種此免疫原之兔子中,三隻在三週後開始產生可用之免疫血清。此免疫血清之Titer及其與AFB1接合之特異性亦由放射免疫法及平衡透析法分別測定,證明其靈敏度可測至每分析試管中0.lng AFB1之量。其他AFB1之代謝物如AFB2,AFG1,AFG2,及AFM1均對AFB1與其免疫血清之結合有不同程度之抑制作用。如經費及時間許可,則此放射免疫分析法可用來大量測定食品之污染程度及探討AFB1與長期肝病之關係。

The feasibility of a specific, precise and quick method for the determination of aflatoxin B1 hasbeen demonstrated. This method is based on a radioimmunoassay procedure developed in this laboratory. Antibodies against aflatoxin B1 were obtained by immunizing New Zealand White rabbits withaflatoxin B1-Poly-L-Lysine conjugate and aflatoxin B1-bovine serum albumin conjugate. An antiserumprepared with the former conjugate was studied in detail. Titer determination and specificity of the serumwere studied using radioimmunoassay procedure and using equilibrium dialysis. At final dilution of 1:1000and 1:4000, it was found that the sensitivity was sufficient to measure 0.1 ng of nonradioactive aflatoxinB1 per assay tube. The specificity of the antibody was also studied by investigating the inhibitions ofaflatoxins B2, G1, G2, and M1 toward the binding of the antibody to aflatoxin B1. The technique isparticularly useful if larger quantities of high titer antisera can be harvested. The method is potentiallyapplicable to routine foodstuff screening and epidemiologic studies.
