- 英文摘要: Glucoamylase could be immobilized on chi-tin and its derivatives-chitosan and N-acetyl-chitosan gel by covalent attachment followedby crosslinking with glutaraldehyde. The op-timum pH, optimum temperature and Km ofchitin-glucoamylase were 4.0, 5.0℃ and 1.3x10-1 g/liter, respectively. It retained 65% ofits activity after storage in a refrigerator forsixty days. Chitosan was a better support for glu-coamylase than chitin. The specific activity ofchitosan-glucoamylase was three times-more thanchitin-glucoamylase and the former had an op-timum temperature of 65℃. Chitosan-gluco-amylase was more stable at an operating tem-perature of 30℃ than at 50℃ in the conversionof soluble starch to glucose. Two types ofreactors had been tested and the result showedthat column type was more convenient andeffective than the batch type reactor. In theoperation of column type reactor, the immobi-lized glucoamylase could retain 60% saccharifica-tion after being used for over one hundred hours.
