顯示幾乎已達均一程度。e - 英文摘要: Using D-tryptophan methyl ester (TME) as a ligand, inexpensive organic polymers chitin, chitosanand polyvinylchloride (PVC) powder as insoluble support, several affinity adsorbents for stem bromelainpurification were prepared. Among the prepared affinity adsorbents, four of them, chitin-ε-amino-n-caproyl-D-tryptophan methyl ester (chitin-ACTME), chitosan-glutaraldehyde-D-tryptophan methyl ester(chitosan-G-TME), chitosan-succinyl-D-tryptophan methyl ester (chitosan-S-TME) and PVC-succinyl-D-tryptophan methyl ester (PVC-S-TME), could strongly adsorb stem bromelain from a mild alkalinesolution and dissociate it in an acidic solution. Stem bromelain was purified from a crude extract by affinity chromatography on a column packedwith chitosan-S-TME affinity adsorbent. By this procedure, the purity of the enzyme increased 2 foldsand the recovery of the activity was 75%. The purified stem bromelain was almost homogeneous as exa-mined by disc polyacrylamide gel electrophoresis.
