觀察受熱壓力(heat stress)作用下大白鼠肌肉的形態學及電鏡細胞結構的變化。本研究以15隻雄性大白鼠為實驗組,
將其固定住後置於攝氏42度溫控箱內使其達到heat stress狀態。另取10隻大白鼠為對照組。所有實驗組大白鼠在受熱處
理後,其四肢明顯無力,同時其血清肌酸磷激素(CK)明顯上升(p<0.01)。將腓腸肌切下,從事組織學,組織化學染色,
電鏡切片,及形態量化分析。結果發現在實驗組的肌肉化學染色中出現明顯的粗糙紅肌纖維(ragged-red fiber),這些纖
維在粒腺體特殊染色的succinate dehydrogenase及細胞色素C氧化?(cytochrome C oxidase)中也同樣呈現強烈反應。再
電鏡形態觀察下,明顯地粒腺體集聚現象,尤其在細胞膜下。經電鏡粒腺體量化分析結果,發現粒腺體的面積在實驗組
的肌細胞中明顯地增加(p<0.001)。這種粒腺體形態及總面積增加的一致性變化發生在受熱壓力後的大白鼠肌細胞內,
意味著這些粒腺體的變化可能是肌細胞在受熱後的啟始的病理現象,而這種現象反應出熱壓力下的肌細胞在能量的代謝
發生了不協調的跡象。
n - 英文摘要: The morphological and ultrastructural alterations of skeletal muscle in experimental rats with heatstress were investigated. Fifteen male Spraque-Dawley rats were exposed in a 42°C constant temperatureoven, resulting in a heat stress state; ten rats were used as controls. All treated rats had weakness of the4 limbs associated with increased serum creatine kinase levels (p<0.01). Soleus muscles were submitted tohistological, histochemical, ultrastructural and quantitative-morphometric analysis. The group receivingheat stress showed many ragged-red fibers in Gomori trichrome stain and appeared hyper-reactive in suc-cinate dehydrogenase and cytochrome C oxidase stains. The ultrastructure of ragged-red fibers showedincreased mitochondrial aggregation as multiple small nests, which were particularly located in the sub-sarcolemmal space. The mitochondrial area was significantly increased in heat stress rats (p<0.001). Theconsistently increased mitochondrial area and histochemical alterations of mitochondria are early patho-logical abnormalities in muscles with heat stress and indicate fundamental impairment of energy metabolism.
