- 中文摘要: 中華黴素A的抗癌作用機轉類似actinomycin D,優先抑制細胞RNA之生合成。中華黴素A與DNA作用的研究結果顯示中華黴素A不能造成DNA的斷裂;但是,可造成該DNA在agarose gel電泳中移動速度的改變。當中華黴素A的濃度逐漸增加時,DNA的移動速度乃逐漸減低;而在達到臨界濃度時,約15.6μg/ml,該DNA之移動速度達到最低程度。此時若再提高中華黴素A的濃度則該DNA的移動速度反而增加。以上之事實顯示中華黴素A與DNA的結合作用為其抗癌作用之主要機轉。
- 英文摘要: The mechanism of action of chounghwamycin A to tumor cells resembles that of actinomycin D,preferentially inhibiting the synthesis of RNA. The interaction of chounghwamycin A with DNA wasstudied, and the results indicated that chounghwamycin A could not cause any scission in tested DNA.However, an induced mobility shift of the DNA in agarose gel electrophoresis was observed. When theconcentration of chounghwamycin A gradually increased, the migration rate of closed circular form DNAis gradually slowed. At a critical concentration of chounghwamycin A, 15.6 μg/ml, the migration rate ofclosed circular form DNA reaches its minimum value. As more chounghwamycin A is added, the mobilityof the closed circular DNA increases gradually again, suggesting that the intercalation of chounghwamycinA in DNA is the primary mechanism of its action against tumor cells.
