- 中文摘要: 新分離之纖維素分解菌六株,業經分別進行形態、培養及生理上各項特徵之鑑定,認為均歸屬於 Cellulomonas 屬菌種。 竹筍殼之成分業經分析。以2% NaOH 溶液於25, 100及121 C下經不同時問前處理竹筍殼後供作基質。以培養微生物。 為欲獲得竹筍殼產製單細胞蛋白質之最佳條件,曾以礦物溶液中加入 CMC 或竹筍殼從事研究不同氮源、不同最初 pH 值、不同基質濃度及不同筍殼∕尿素比對菌株之影響。為欲提高培養基中基質之濃度,曾以部分尿素及竹筍殼於菌株培養期問追加,發現其結果較二者於最初配製培養基時全部加入者所得之結果為高。以 500 ml 三角瓶中含 50 ml 培養基所得之最佳結果為450 mg 蛋白質,相當約每公升中含9克蛋白質“以6公升之醱酵槽內含3公升3%基質之培養基於30 C下培養48小時,可獲得每公升含 41.5%蛋白質之乾物質12克,但在6%基質時,可獲得25克僅含 25.6%蛋白質之最後產品,故對以醱酵槽培養菌體,其最佳培養條件尚待進一步之研究。
- 英文摘要: The morphological, cultural and physio-logical characteristics of six new isolates ofcellulolytic bacteria were fully studied and allthe organisms were identified as members ofthe genus Cellulomonas. The chemical composition of the husk wasanalyzed. Husk pretreated with 2% NaOHsolution under 25, 100 and 121 C for varioustimes respectively were used for cultivation ofthe organisms. In order to obtain optimalconditions for the production of SCP from thepretreated husk, the mineral solution containingCMC or husk has been used to study the effectsof nitrogen source, initial pH value, substrateconcentration and husk/urea ratio on the growthof the organisms. To increase the concentrationof the substrate in the medium, supplementaladdition of urea and pretreated husk respectivelyduring the growth of the organisms has beencarried out and better yields were obtained incomparison with both components added into themedium at the very beginning of the experiment.The maximal SCP produced was around 450mgper 50ml medium in a 500ml shaken flask,which is equivalent to 9gm protein per liter.Twelve gm per liter of dried product containing41.5% protein were obtained from 3 liter ofmedium with a 3% substrate in a 6 liter-fer-mentor after 48 hours cultivation at 30 C.However, 25gm of the final product containingonly 25.6% protein per liter were harvested inthe same fermentor with a 6% substrate. Theoptimal conditions for the cultivation of theorganisms in fermentor need to be furtherstudied.
