- 中文摘要: 本文是以免疫酵素法(immunoperoxidase-antiperoxidase, PAP)來證實在單層培養中的星狀神經膠細胞株含有GFAP的存在,而這些細胞株乃由新生正常大白鼠的腦組織細胞經過3年的繼代培養而獲得。 從典型的PAP染色法吾人可觀察到GFAP緊密地結連在星狀神經膠細胞的細胞質細纖維構造中。從這些細纖維的分佈與外貌乃可判斷為星狀神經膠細讖維,他們經常聚集在細胞之胞突與核周邊之胞質中。 本研究結果有力地暗示(1)吾人可長期培養星狀神經膠細胞株而並不致失去他們之分化特徵,即仍然可以保持形成與貯存GFAP的能力,(2)這群星狀神經膠細胞可以為吾人將來從事該群細胞的行為和機能方面的研究提供極具利用價值的細胞種型。
- 英文摘要: Glial fibrillary acidic protein (GFAP) was demonstrated by indirect immunocytochemical (PAP)method in the monolayer cultures of astrocyte-like cell line derived from normal new-born rat brain tissuecells and established through the routine continuous passaging procedures for about 3 years. It was demonstrated that the characteristic staining of GFAP is closely associated with intracyto-plasmic fibrillar structure of these cells. From the distribution and appearance they could be considered asastrocytic filaments. They were concentrated in the cytoplasmic processes and perinuclear area. These results strongly suggest that (1) it is possible to maintain the astrocyte-like cell line for long-term cultures without losing their differentiated characteristics, namely the ability to synthesize and pre-serve GFAP, (2) our astrocyte cultures will serve as a very useful model for future studies of the behavioursand functions of astrocytes.
