- 英文摘要: Chronic and excessive ethanol consumption is known toalter neuron and glial cell functions in the central ner-vous system (CNS). Astrocytes comprise the major celltype in the brain. These immune active cells are capableof responding to proinflammatory cytokines and endo-toxins, which stimulate transcriptional pathways leadingto induction of genes, including the inducible nitric ox-ide synthase (iNOS) and secretory phospholipase A (sPLA ). In this study, we investigate the effects of etha-nol on cytokine-induced iNOS and sPLA in immortalizedastrocytes (DITNC). When DITNC cells were exposed toethanol (0-200 mM) for 4 h prior to subsequent stimula-tion with cytokines for 16 h, NO production decreasedwith increasing ethanol concentrations starting from50 mM. At ethanol concentrations higher than 100 mM,ethanol also inhibited cytokine-induced sPLA releaseinto the culture medium. The inhibitory effect of ethanolon NO production corresponds well with the decrease iniNOS protein and NOS enzyme activity, but not withiNOS and sPLA mRNA nor binding of NF- B to DNA.The inhibition of cytokine-induced NO production by eth-anol was also dependent on the time of ethanol expo-sure to the cells, but addition of acetaldehyde up to200 M did not elicit any changes. Taken together, theseresults provide evidence for a posttranscriptional modeof ethanol action on the cytokine induction pathway forNO production in astrocytes.
