- 中文摘要: 本研究以動力學解析方式探討脯胺酸與異白胺酸對 Pseudomonas amyloderamosa JD210 在 isoamylase 生產上之影響效應。在批式培養中,菌體之生長可以很適切地以 logistic 模式來表示。酵素之生產係遵循生長關聯型態,其生產狀況可用Luedeking-Piret模式來描述。再者,培養基中的麥芽糖、脯胺酸及異白胺酸等基質之消耗情形,則可成功地以修飾型Luedeking-Piret模式來表示。 基於動力學解析之結果顯示,在合成培養基中,脯胺酸與異白服酸的最適濃度分別為0.15%及0.036%。而在低於此最適濃度值以下時,這兩種胺基酸對於酵素生產速率呈現了明顯的加成效應。
- 英文摘要: The production of isoamylase by Pseudomonas amyloderamosa JD210 was investigated kinetically.The cell growth in a batch culture was found to be successfully expressed by a logistic model. Enzymeproduction followed the growth-associated pattern, and the Luedeking-Piret model could be used to describethe data properly. Moreover, the substrate utilization of maltose, proline and isoleucine were also satis-factorily represented by a modified Luedeking-Piret model. The effects of proline and isoleucine on enzyme production were studied based on the variation ofkinetic parameters. The optimal concentrations of proline and isoleucine were found to be 0.15% and0.036%, respectively. Furthermore, a significant additive effect of these two amino acids on enzymeproduction rate was also observed.
