- 中文摘要: 本文提出一個數學模式去描述蛋白質由聚乙醯胺膠質電泳轉漬到薄膜的過程。實驗顯示這個過程是不可逆的,而且對某一種蛋白質而言,初期的轉漬效率與該蛋白質在膠質的濃度無關,在某一狀況下是個定值。因此在此時期蛋白質電泳轉漬與一級不可逆反應類似;此一過程的反應常數可以由膠質中蛋白質在最初30分鐘減少的量計算得知。因此各種變數諸如蛋白質分子大小,電流,溫度,轉漬緩衝液中甲醇的濃度以及不同薄膜對反應常數的影響都被用來檢測此一數學模式的正確性,實驗結果基本上與假設一致。
- 英文摘要: A mathematical model has been proposed from this study to describe the electrotransfer of proteinfrom polyacrylamide gels to membranes. The process of protein blotting has been shown here to be ir-reversible and the initial transfer efficiency to be constant with different amounts of protein. Thus, theprotein transfer process during the initial stage is similar to a first order, irreversible reaction. The initialrate constant of the process was calculated by measuring the elution of protein from the gel in the first 30min.The effects on the rate constant due to factors such as protein size, current, temperature, methanolconcentration in the transfer buffer and membrane have been studied to test the proposal. The results arevirtually in accordance with the hypothesis.
