抗胺基酸類似物突變水稻的選拔及遺傳

以含各種胺基酸類似物的分化培養基培養水稻的小孢子癒合組織,以選出抗性細胞及植株。在含5-甲基色胺酸的分化培養基上得到四棵綠苗及一個細胞系;在含乙硫胺酸的分化培養基上得到一棵綠苗及二個細胞系;在含對氟苯初油胺酸的分化培養基上得到一個細胞系。在這些植株及細胞系中,除了發現一棵植株的體細胞癒合組織對5-甲基色胺酸不具抗性外,尚未證實其餘植株及細胞系對相關的胺基酸類似物是否仍保持抗性。不過對於以前得到的三棵抗5-甲基色胺酸的植株的抗性已有更深入的研究。在這三棵植株中,除了一棵已死亡外,其餘二棵的體細胞癒合組織對5-甲基色胺酸仍具抗性,顯示他們是抗5-甲基色胺酸的突變體。它們自交後代的體細胞癒合組織對5-甲基色胺酸的抗性分為強抗、中抗及不抗三種,顯示控制此種抗性的為一半顯性基因。

Microspore callus of rice (Oryza sativa L.)was cultured on a medium containing the aminoacid analog 5-methyltryptophan (5-MT), ethi-onine (Eth), or parafluorophenylalanine (PFP).A few cell lines survived and some of themdifferentiated into plants. One plant selectedfrom callus cultured on the 5-MT-containingmedium was later found to be nonresistant tothe analog. The resistibility of the other plantsand cell lines to the respective analogs has notbeen tested. Inheritance of the resistant trait oftwo 5-MT-resistant plants obtained last year wasstudied. It was observed that callus initiatedfrom the somatic tissues of their self-pollinatedprogeny plants could be classified into threedistinct groups in response to the analog: highlyresistant, moderately resistant, and nonresistant.This result suggests that the resistant trait isprobably determined by a semidominant gene.
