- 中文摘要: 著者等從土壤中分離篩選出能產生澱粉水解酵素抑制劑之優良菌株 NTU-3314 號菌,此菌株在菌學上已被鑑定為Streptomyces nigrifaciens NTU-3314。 將此菌株預先培養於種培養基後,以接種量為15%接種於含有 defatted soybean flake 3.0%o, potato starch 4.0%,casein 0.6%, sucrose 0.6%, serine 0.02% 及NaCl 0.8% (pH 7.0)之生產用培養基2公升之5公升醱酵槽中,在培養溫度30°C,通氣量 1.5vvm 及攪拌速率 300rpm 下進行通氣攪拌培養,經培養24小時後,可以得最高之酵素抑制劑產量。 此酵素抑制劑對α-amylase和 glucoamylase 具有抑制作用,但對 β-amylase, α-glucosidase, β-glucosidase 及 dextra-nase則無抑制活性。所得酵素抑制劑在O.1M磷酸緩衝液(pH 7.0)中非常安定,即使在100°C之沸水中加熱20分鐘,仍能完全保持其活性。
- 英文摘要: In the course of screening amylase inhibitor producing, microorganisms, a strain identified as Strep-tomyces nigrifaciens NTU-3314 was found to have the highest inhibitor-producing ability among the otherisolated strains. This strain was aerobically cultured at 30°C in a 5 jar fermentor with a working volume of 2 .The optimum cultural medium consisted of defatted soybean flake 3.0%, potato starch 4.0%, casein 0.6%,sucrose 0.6%, serine 0.02% and NaCl 0.8% (pH 7.0). With an aeration rate of 1.5 vvm, an agitation speedof 300 rpm and an inoculum of 15% seed (previously grown in seed medium 3), the highest amount ofinhibitor was obtained after 24 hours of cultivation. The amylase inhibitor produced had inhibitory effects on both α-amylase and glucoamylase, but noton β-amylase, α-glucosidase, β-glucosidase or dextranase. It was quite stable in O.1M phosphate buffer(pH 7.0) ana nearly 100% of its activity was retained even alter bolllngat 100°C for 20 min.
