- 中文摘要: 由本省家蠶所分離之多角體,以0.05M NaCl+0.008M Na2CO3 (pH10.5)鹼性溶液處理,未溶部份以低速離心沉 澱,再將此沉澱物以0.025M NaCl + 0.08 M Na2 Co3 (pHll)之鹼性溶液處理,所得病毒粒子經電子顯微鏡觀察之結 果,均?桿狀物,大小約70 - 80X300 - 350nm ,由此顯示此種家蠶多角體係核多角體病毒(nuclear polyhedrosis virus, NPV)。進一?之純化發現採用20—40% (W/V)蔗糖梯度溶液在57,700g離心40分鐘可得三條分離之白色帶狀物,經電 子顯微鏡觀察,最上一條?核多角體病毒之包被粒子及裸露病毒粒子,第二條及第三條則完全?裸露病毒粒子所組成。 此法可取得大量的純化完整病毒粒子,可供本省將來純化此病毒之參考,並可供與此病毒相關的病理學及血清學研究 之基礎。
- 英文摘要: Virus pellet containing enveloped virions was obtained by treating the polyhedra isolated from the silkworm, Bombyx mori, reared in Taiwan, first with 0.05 M NaCl + 0.008 M Na2C03 (pH 10.5) for 5 min, followed by 0.025 M NaCl + 0.08 M Na2COa (pH 11) for 1 hr, and then by centrifuging at 40,000 g for 30 min. Electron micrographs showed that the pellet contains numerous rod-shaped virions, measuring ca. 70-80 x 300-350 nm. This observation revealed that the polydedrosis virus infecting the silkworms of Taiwan was a type of nuclear polyhedrosis virus (NPV). Further purification of the virus particles was found satisfactory by ultracentrifugation at 57,700 g for 40 min. through a 20-40% (w/v) sucrose gradient. Three white bands, which contain either enveloped virions and naked virus particles or naked virus particles only, were formed in the gradient, providing a useful technique for purifying the silkworm NPV.
