- 英文摘要: tured smooth muscle might be used as a model to studythe regulation of contractile function of smooth muscleby various signal transduction pathways.Smooth muscle cells lose their contractile function andphenotype very rapidly when placed in culture. Duringorgan culture of smooth muscle strips, phenotype is lostmore slowly. In the present studies, we established anorgan culture model to study contractile function andexpression of muscarinic receptors, G proteins and ade-nylyl cyclase in different serum concentrations in tra-cheal smooth muscle from swine. The results show thatcontractile function and the amounts of M receptors, Gproteins and adenylyl cyclase were maintained for up to5 days in culture. The expression of M receptors wassignificantly decreased in culture when compared tofreshly isolated muscles. Maximal isometric tension wassignificantly increased in cultured muscles comparedwith freshly isolated muscles. Different serum concentra-tions did not significantly affect contractile function andexpression of muscarinic receptors, G proteins and ade-nylyl cyclase. In conclusion, our studies suggest that cul-
